RiboCleave™ 1
Sequence-specific RNA restriction enzyme
RiboCleave™ 1
OverView
RiboCleave™ 1 is a sequence-specific RNA restriction enzyme that recognizes and cleaves the GAA↓AU sequence in single-stranded RNA. Designed for RNA analytics applications, it enables rapid generation of defined analytical fragments for mRNA cap-efficiency assessment. A single digestion step replaces complex multi-step RNase H workflows, reducing sample preparation time from approximately 6 hours to less than 30 minutes. As a sample-preparation step, RiboCleave™ 1 is method-independent by design: the same digest can be analyzed by urea-PAGE, Bioanalyzer or LC-MS.
Key Benefits
- Reduces cap-analysis sample preparation from approximately 6 hours to less than 30 minutes.
- Eliminates guide oligo design and optimization requirements.
- Method-independent by design: one digest for urea-PAGE, Bioanalyzer and LC-MS readouts.
- Enables in-house cap analysis on standard gel equipment.
Key Features
- Sequence-specific cleavage of GAA↓AU motifs in single-stranded RNA.
- Single-enzyme, one-incubation workflow.
- Compatible with natural and modified RNA substrates, including N1-methylpseudouridine and pseudouridine.
- Supplied with dedicated Reaction Buffers for analytical flexibility on gel and/or HPLC/LC-MS readouts

FAQs
RiboCleave™ 1 is a sequence-specific RNA restriction enzyme that recognizes the GAA↓AU sequence in single-stranded RNA and generates defined cleavage products suitable for downstream analytical workflows. It also exhibits star activity on the following sequences: AAA.AU ; GAA.AA; GAA.AC ; GUAAU. Other star-activity sequences may exist.
RiboCleave™ 1 performs optimally between 30-40°C, pH 7.7-9.0, and 20-60 mM NaCl. Activity is highest when less than 0.5 mM Mg²⁺ is present, although the enzyme remains active at Mg²⁺ concentrations up to 6 mM.
Yes. RiboCleave™ 1 has been demonstrated to cleave RNA containing therapeutically relevant modified uridines, including N1-methylpseudouridine (m1Ψ) and pseudouridine (Ψ), enabling its use in modified mRNA analytical workflows.
No. Unlike RNase H-based cap-analysis workflows, RiboCleave™ 1 does not require guide oligonucleotide design, hybridization, or optimization. A single enzyme digestion step produces analytical fragments that are ready for downstream analysis.
RiboCleave™ 1 is designed as a method-independent sample-preparation step. It has been tested with urea-PAGE, Bioanalyzer and LC-MS. If you’re planning to use another analytical method, contact us to find out whether supporting data is available.
