‍

RiboClean™ dsDNase is a double-strand specific DNase designed for effective removal of template DNA from in vitro transcription (IVT) reactions. The enzyme selectively degrades double-stranded DNA while preserving RNA integrity and yield, enabling streamlined RNA manufacturing workflows with less DNase protein going into purification. Under tested IVT conditions, RiboClean™ dsDNase achieved template DNA digestion using up to 10× fewer enzyme units than conventional DNase I.

 Key Benefits

  • Removes template DNA with up to 10x fewer enzyme units than DNase I.
  • Reduces residual template DNA while maintaining RNA integrity and yield.
  • Allows 40x less DNase protein by mass than DNase I, reducing DNase carry-over into purification.
  • Integrates directly into IVT workflows without additional calcium supplementation or buffer exchange.
  • Modeled >80% lower DNase-related cost per 1g batch, Modeled >10% lower reagent cost per mg RNA produced.

Key Features

  • Generates DNA fragments <17 nt.
  • Optimal activity between 35–45°C.
  • Active across a broad pH range and tolerant to variable process conditions.
