M-SAN HQ - Optimum Depletion of Host Cells DNA
M-SAN HQ is selected for workflows that demand reliable and effiecient host DNA depletion without compromising sensitive pathogen detection, including RNA viruses. It works directly in minimally processed samples such as plasma, swabs, or urine at physiological salt concentrations (125 to 250 mM NaCl).
In some cases, high concentrations of salt can reduce recovery of fragile pathogens like enveloped RNA viruses. M SAN HQ is exceptionally active at physiological salt levels, making it the obvious choice when the goal is to detect and analyse both bacterial and viral DNA/RNA from the same sample. This enables unified RNA and DNA detection from a single sample. No prior purification is needed.
When supplemented with low amounts of Magnesium, M-SAN HQ works efficiently directly in most patient samples (physiological salt and pH) without the need for buffer adjustments.

M-SAN HQ has optimal performance in the 7.2 - 8.7 pH range. This allows very high performance in most biological buffers. With a working range starting at pH 6.5, M-SAN HQ is also a candidate for DNA removal from insect cell media. The activity was measured at 37°C in a 25 mM Tris-HCl or Bis-Tris buffer at various pH, 2.5 mM MgCl2 and 150 mM NaCl. Activity measured at pH 7.7 was set to 100%.

M-SAN HQ is highly efficient for digesting DNA at 37°C, and tolerate temperatures up to 50°C. The activity was measured in a 25 mM Tris-HCl buffer at pH 7.8 (@measurement temperature), 5 mM MgCl2 and 150 mM of NaCl. 37°C was set to 100% as this is the temperature used for defining the activity in Units.

Yes. M-SAN HQ can be used in workflows targeting both DNA and RNA pathogens. During host-DNA depletion, microbial and viral genomes remain protected within their intact cells or viral capsids, allowing efficient removal of host DNA without compromising detection or sequencing of pathogen DNA or RNA in downstream assays.
M-SAN HQ shows peak nuclease activity under physiological conditions—typically 125–250 mM NaCl, pH 7.2–8.7, and 36–50 °C. An Mg²⁺ concentration of 4–15 mM supports optimal activity.
These conditions align closely with clinical and biological sample buffers, enabling efficient host-DNA removal within standard diagnostic workflows.
M-SAN HQ functions in a wide range of physiological and diagnostic buffers used for clinical and biological samples such as swabs, respiratory fluids, and liquid biopsies. A minimum of 0.5 mM Mg²⁺ is required for activity.
The enzyme remains active in moderate to high-salt and standard sample-preparation buffers, allowing direct use for host-DNA depletion without changing existing workflows.
M-SAN HQ can be effectively inactivated by reducing agents or digested using a proteinase.
